Prevalence of Candida albicans in patients using fixed orthodontic appliances after the use of peppermint

Abstract

This study was conducted to evaluate the antifungal effectof peppermint extract as mouth rinse during orthodontic treatment.Thirty patients (11 males and 19 females) who wear orthodonticappliances and instructed to use chlorhexidine asmouth rinse (8 patients); Mentha piperita extract (10 patients)or left to care of their teeth without any mouth rinse (12 patients);mean number of Candida albicans colonies recoveredfrom anterior surface at three intervals (Time 0: Before wearingthe appliance; time 1: After 7 days and time 2: After 1month) was 48.8, 144.3, 158.9 for the control group; 132.9,125.4, 147.9 for the peppermint group and 72, 105.8 and130.8 for the chlorhexidine group.The mean number of Candida albicans colonies fromthe posterior surface at the different intervals was 87.4, 136.8,159.5; 121.3, 84, 34.6 and 78.5, 91.25, 91 for control, peppermintand chlorhexidine groups respectively. The use of thepeppermint extract decreases the number of Candida albicanssignificantly at the posterior surfaces

The Effect of Peppermint Extract (Muntha piperita)and Incubation Period on the Growth of Some Plant Pathogenic Fungi

Abstract

The current research aims at studying the effect of the aqueous extract of mint plant (Muntha piperita) on growth of some fungi such as ; Rhizoctonia solani, Aspergillus niger, Aspergillus terreus, Fusarium oxysporum, Fusarium oxysporum f.sp. The extract has been added to the PDA media at four concentrations 25%, 50%, 75% and 100% in addition to the control treatment (standard PDA media only ) . The results show the effectiveness of the aqueous extracts of mint plant in the inhibition of fungal growth . The highest inhibition is at 100% concentration of the extract , at which the growth of Rhizoctonia solani recorded values of colony diameter; 4.49,6.83, and 8.33 cm for the incubation periods of 1,2,or 3 days , respectively , in comparison with Aspergillus terrens which records the lowest growth values ; 1.82, 2.55, and 3.93 cm colony diameter for the same incubation period as above , respectively . However , the fungus Rhizoctonia solani is not affected by the extract treatments, especially after the incubation periods of 4,5,6,or 7 days . The results also show significant effect of the media on fungal growth . The standard PDA medium gives the highest fungal growth values of colony diameter ; 3.65, 49.13, 5.85, 6.64, 7.23, and 7.72 cm for all incubation periods in compare with 100% concentration of the extract which records values of colony diameter; 3.71, 5.07, 5.89, and 6.64 cm for the same incubation periods , respectively . For the interaction , there is a significant effect of the combination of Fusarium oxysporum and 100% concentration of the extract for the incubation period of 1,2,and 3 days , while the combination of Aspergillus niger , and 100% concentration gave significant effect for the incubation period of 4,5,6, and 7 days in comparison with to other treatment .

يهدف البحث الى دراسة تأثير مستخلصات نبات النعناع (Muntha piperita ) المائية في نمو فطريات (Rhizoctonia solani , Aspergillus niger , Aspergillus terreus , Fusarium oxysporum , Fusarium oxysporum f.sp. lycopersici) اذ اضيف مستخلص النعناع بعدة تراكيز الى الوسط PDA (25%,50%, 75%, 100%) وكانت معاملة السيطرة للوسط PDA القياسي . اضهرت النتائج فعالية المستخلصات المائية لنبات النعناع في تثبيط نمو الفطريات وكان اعلى تثبيط عند استخدام التركيز 100 % . اذ بلغ النمو القطري للفطر Rhizoctonia Solani ( 8.33, 6.83 , 4.79 ) سم . لفترات الحضن 3,2,1 يوم على التوالي مقارنة بلفطر Aspergillus terrens والذي اعطى اقل نمو قطري اذ بلغ (3.39, 2.55, 1.82) سم لنفس فترات الحضن وعلى التوالي . في حين لم يتاثر الفطر Rhizoctonia Solani بالمستخلص لاسيما بعد فترات الحضن 7,6,5,4 يوم . كما بينت النتائج تاثيرا معنويا للوسط في النمو القطري للفطريات اذ اعطى الوسط ( PDA القياسي) اعلى نمو بلغ (.72 , 7.23, 6.64, 5.85, 4.9 3.65 ) سم ولجميع فترات الحضن مقارنة بالتركيز 100% الذي اعطى ( 6.64, 5.89, 5.07 , 3.71 ) سم لنفس فترات الحضن على التوالي واظهرت نتائج التداخل معنوية في المعاملة (Fusarium Oxysporum * 100% ) لفترات الحضن 3,2,1 يوم في حين اعطت معاملة Aspergillus niger * 100 % تداخلا معنويا . مقارنة بالمعاملات الاخرى ولفترات الحضن 7,6,5,4 يوم .

Abstract

Background Due to the increase in the microorganism resistance to most of conventional antifungal drug, studies have encouraged to discover novel treatments for infections caused by Candida species. Hence, alternative therapies are crucial and the use of herbal medicines seems being a promising solution for the candidiasis treatment. Materials and Methods the antifungal effect of some medicinal plant extracts (peppermint and pomegranate) was confirmed by some studies. But there was still a lack of knowledge associated with the specificity of these extracts on sensitivity and viability counts of candida albicans, so for this reason this study was conducted. Water extract (peppermint and pomegranate) was prepared, different concentration (5, 10, 15, 20, and 25mg/ml) from them were taken, tested candida albicans was used to evaluate these extracts on its sensitivity, viability counts, and minimum inhibitory concentration (MIC) (in vitro). Results showed that all concentration of pomegranate showed zone of inhibition, there were highly significant differences between concentrations except between (10, 15) and (20, 25), but peppermint revealed zero zone of inhibition on Sabouraud dextrose agar medium. All concentrations of both extracts were effect in reducing viability counts of candida albicans with highly significant differences with Negative control (Distilled water) P<0.05, but there were no significant difference P >0.05with positive control (Nystatin) and with each other. Conclusions All plant extracts used in this study had antifungal effect and influence on candida albicans. For this reason can be used as disinfectants, mouth wash or soaking solutions and as an alternate to drug in the treatment of denture stomatitis.
