Abstract

The present study was carried out to determine the effect of super benzene inhalathion and its metabolites on haemopoietic tissues(spleen and bone marrow) on experimental rats.Animals were exposed via inhalation to concentrations of (5 and 10) ppm super benzene for 2 hour/day, 5 days /week for 3 months.Histological studies revealed that spleen of the exposed animals to super benzene, were exposed to 5 ppm showed white pulp atrophy and absence of megakaryocytes in red pulp with blood vessels congestion.In 10 ppm showed increased of extramedulary haemopoiesis, acute splenitis and fibrosis as well as showed aggregation of lymphocytes in lymph node, thickness with centeral artery wall also inflammation of inflammatory cells inside centeral artery and vasodilatation in centeral artery in asplenic nodule.While the bone marrow related to the animals exposed to 5 and 10 ppm showed reduced in haemapoietic tissue which partly replaced by adipose tissue.

تمت دراسة تأثير أستنشاق بخار البنزين المحسن على الانسجة المكونة للدم ( الطحال ونقي العظم) والمستخدم حاليا كوقود للسيارات ومولدات التوليد الكهربائية في الجرذان المختبرية. عرضت الحيوانات الى التراكيز ( 5 و10 ) جزء بالمليون من بخار مادة البنزين المحسن وبمعدل 2 ساعة / يوم و 5 يوم/ اسبوع ولمدة 12 اسبوع. اظهرت الدراسة تغيرات نسجية مرضية في الطحال تشتمل على ضمور اللب الابيض واحتقان للاوعية الدموية اضافة الى ظهور حالة التليف وزيادة في سمك جدار الشريان المركزي مع ارتشاح خلايا دفاعية داخل الشريان الطحالي. كما اظهرت الدراسة نقصان في خلايا الدم الناتج عن خلل وظيفي في نقي العظم مما ادى الى احلال النسيج الدهني بدل الخلايا . أكدت النتائج الحالية على التأثيرات السلبية لبخار هذه المادة من خلال التغيرات المرضية في تركيب نسيج الطحال ونقي العظم .

Abstract

In an assessment of cell mediated immunity against Salmonefl g_'p_h_i antigen inGuinea pigs following their intraperitoneal injection with sensitized spleen cells. Theresults were revealed a high level of cell mediated immunity against Salmonella typh_i_antigen in the Guinea pigs intraperitonealy received the sensitized spleen cells (5x1 08/1111).The level of cell mediated immunity was detected using delayed type hypersensitivity test.Macrophages migration inhibition test and Erythrocytes rosette test.The nonnal (nonsensitized) spleen cell recipient group (control group) was showed nolevel ofcell mediated immunity using these tests.

Abstract

We presented here a 25years old lady with a multiple hatdtid cysts in lung and liver and spleen in large number in each organ. Which regarded inoperable and difficult to treared.We use medical treatment with antihelimenthes drugs in new regime and follow-up over 1year clinically, laboratory investigations, U/s and CT.scan. There was good disappearance of liver and spleen cysts with very good response in lungs.

Abstract

A total of 20 female mice (8 week of age)a 10 mice control and 10 deal with 10¬¬¬5cfu per animal of¬¬¬ Brucella melitensis by injection intraperitonial. Samples were collected over a 6 week period of infection. Blood collected for serological test revealed positive for animals were injected with bacteria then animal dissected, liver and spleen collected and kept in 10% formalin for histological study.Results showed Histopathological changes show congestion, granulomatous, fatty change,fibrosis in liver and increased number of lymphohistiocytic cells and increased amount of white pulp. The study concluded thatthepathological changes in liver and spleen caused by brucellamelitensis in mice are similar to those observed in humans with brucellosis

تناول البحث دراسة تأثير الإصابة ببكتريا البروسيلا من جنس Brucellamilitensis ودرجة الضراوة لها على أنسجة كلا من الكبد والطحال فيالفئران.تضمنت الدراسة استخدام 20 من الفئران الإناث بعمر ثمانية أسابيع, 10 منها استخدمت مجموعة سيطرة و10 أخرى تمت معاملتها ببكتريا البروسلا حيث تم حقن 10¬¬¬5من عزلة نقية للبروسيلا في البريتون. حضنت البكتريا بالحيوانات لمدة ثمانية أسابيع بعدها تم تخدير الحيوانات وجمع عينات الدم للفحوصات السيريولوجية ثم شرحت الحيوانات وجمعت الأعضاء المنتخبة وتم تحضيرها الدراسة النسيجية . أظهرت النتائج وجود تغيرات واضحة في أنسجة كل من الكبد والطحال تمثلت بظهور احتقان وتغيرات دهنية وتلييفات وغيره في الكبد مع تنخر وزيادة نسبة اللب الاحمر في الطحال مع تجمع للخلايا الالتهابية.

Abstract

Water extract of castor bean was prepared and injected in mice in order to study the effect of acute phase of toxicity (in different doses) on living tissue, for these reasons, 24 male white mice were divided equally into 4 groups. The 1st, 2nd, and 3rd groups were injected with the aqueous extraction of the castor seeds subcutaneously with 20, 30 and 60 mg/Kg of body weight, respectively, while the 4th group used as control group. After 24 hrs. the internal organs showed degenerative changes and proteintious material in the spleen and kidney, also these organs in addition to liver and stomach showed inflammatory reaction in their parenchyma. The lesions in the examined internal organs were mild in a dose of 20 mg/Kg B.W. and more sever lesions seen in a dose of 60 mg/kg of body weight.

Abstract

Background: The spleen is the most common solid organ injured in patients who had sustained abdominal trauma. Such injuries to the spleen represent approximately one quarter of all blunt injuries of the abdominal viscera.Due to its remarkable vasculature and its fragile structure, splenic rupture is the most widespread cause of intra-abdominal hemorrhage.Objective: To assess the magnitude of splenic injury, the management of splenic injury, and to evaluate the postoperative complications.Methods: A prospective study of 57 cases of splenicinjury was performed in Al-Kadhimiya TeachingHospital during the period between the 1st of October2004 and the 1st of October 2006. Statistical analysiswas then performed to identify the causes, managementand postoperative complications. Splenic injuries weregraded into 5 grades.Results: The highest incidence of splenic injury was recorded in age group s31-40 year. Penetrating injury was much more common than blunt trauma (73.68% versus 26.31%).Fifty patients (87 %) were diagnosed by exploratory laparotomy, ultrasonography in 5 patients (8.77 %), CT scan was positive in 2 patients (3.5 %). Associated injuries were recorded in 41 patients 71.92%. Pulmonary complications were the most frequent complications (18 patients, 31.57 %).Conclusion: Splenic injuries are common surgical problems in Al-Kadhimiya Teaching Hospital. Still there are many difficulties in establishing the diagnosis of the splenic injury with shortage in the new diagnostic modalities especially (CT -Scan, MR1 and even U/S scan) in the emergency situations.In spite of the dangerous post-operative complications of splenectomy, it is still the most commonly performed surgical procedure in cases of splenic injury.

Abstract

The present study was conducted to determine the histological and Molecularchanges in the spleen of adult male Japanese quail after being exposed to effect ofthermal stress was recorded. This study included two groups, each group consistingtwelve male birds, the control group (A) was exposed to normal temperature for45days, while the group (B) exposed to temperature (42OC) for 45 days. Thehistological and molecular changes were studied during (15, 30 and 45) days of theexperiment, histological changes in spleen were represented by necrosis,degeneration hemorrhage and shrinkage fibrosis of lymphocytic nodules,cytoplasmic vaculation, infiltration of lymphocytes, metamorphosis of lymphocyte,nuclei pyknotic, dilation in the central vein of lymphatic nodules, edema, dilation ofsinusoids and congested blood vessels. Molecular examination showed that the heatshock protein (hsp70) gen is present in temperature group that is also found in thecontrol group. This confirms that the hsp70 gen is present in birds at normal andabnormal conditions.

Abstract

ABSTRACT
Murine heat shock protein 90 beta isotype (84kDa) was purified from the
cytoplasm of BALB/c mice spleen cells. Since this protein dose not have an in vitro
activity assay and the presence of protein with molecular weight equal or near 84 kDa
was followed in each purification step. Cytoplasm fractions of spleen cells contained
proteins of molecular weight (MW) ranged from 86.0 to 20.9kDa as determined with
SDS-PAGE analysis. The ammonium sulfate 70% saturation precipitate of cytoplasm
fraction exhibited protein bands with molecular weight ranged from 40.7 to 86.1kDa.
Ion exchange chromatography with DEAE-Cellulose column for the ammonium
sulfate precipitated cytoplasm proteins produced one prominent peak, eluted between
0.5 and 0.7M NaCl. This peak contain four proteins with MW ranged from 19 to
83.1kDa. During gel filtration through Sepharose 6B column for the peak eluted from
DEAE-cellulose column, a peak with estimated molecular weight of 87kDa was
selected among others produced. Second round of gel filtration through Sephacryl S-
200 was conducted for the 87kDa selected fractions eluted from the previous gel
filtration step. The presence of one peak was emphasized and its estimated MW was
83.176 kDa. The molecular weight and purity of this protein was checked again with
SDS-PAGE 10%. The identity of the purified proteins as Hsp90&#946; was confirmed with
immunofixation assay by gel electrophoresis on 1%agarose using standard.

الخلاصةتمت تنقية بروتين الصدمة الحرارية 90 نمط بيتا ( 84 كيلودالتن) من سايتوبلازم خلايا طحال الفئران البيضوبسبب عدم وجود فحص فعالية خاص بالكشف عن هذا البروتين فقد أعتمد وزنه الجزيئي .BALB/c ضربكأحد الأدلة على وجوده خلال مختلف مراحل التنقية. أظهر الترحيل الكهربائي بإستعمال الاكرل امايد المتعددلجزء ساتوبلازم خلايا الطحال وجود حزم بروتينية تراوح وزنها الجزيئي (SDS) بوجود المادة الماسخة للبروتينبين 20.9 و 86 كيلودالتن. رسب ذلك الجزء بوساطة ملح كبريتات الامونيوم بنسبة إشباع 70 % وظهر أنهيحتوي حزم بروتينية تراوح وزنها الجزيئي بين 40.7 و 86.1 كيلودالتون. أظهرت نتائج خطوة التبادل الايونيللبروتينات المترسبة بكبريتات (DEAE-Cellulose) بإستعمال المبادل الآيوني ثنائي اثيل امينواثيل سليلوز0.7 مولرمن كلوريد الصوديوم و إحتوت – الامونيوم وجود قمة واحدة أستردت بتدرج ملحي تراوح بين 0.583.1 كيلودالتون. جرت عملية الترشيح الهلامي – أربعة حزم بروتينية تراوح الوزن الجزيئي لها بين 19للقمة المستردة من خطوة التبادل الآيوني وجرى إسترداد عدة قمم من العمود Sephacryl 6B بإستعمال هلام84 كيلودالتون ) ) وأنتخبت قمة واحدة تمثل وزن جزيئي قريب من الوزن الجزيئي لبروتين الصدمة الحرارية 90هي قمة بوزن 87 كيلودالتون. أعيدت خطوة الترشيح الهلامي للقمة المنتخبه من الخطوة السابقة وأستعمل هذهأكدت خطوة الترشيح الهلامي الثانية وجود قمة بروتينية واحدة منفردة في .Sephacryl S- المرة هلام 200الأجزاء التي تم جمعها من خطوة الترشيح الهلامي الأولى، وأن تلك القمة تمتلك وزن جزيئي 83.176كيلودالتون. جرى التأكد من نقاوة ووزن تلك القمة بالترحيل الكهربائي على هلام الاكريل امايد المتعدد وبوجودوكانت النتيجة أن هناك حزمة بروتينية واحدة ذات وزن 83 كيلو دالتون . تم (SDS) المواد الماسخة للبروتينالفأري بإستعمال فحص التثبيت المناعي بالترحيل Hsp90β التحقق من هوية تلك الجزيئة البروتينية على إنها(monoclonal الكهربائي على هلام الاكاروز 1% وبتوظيف عدة تشخيص ذات أضداد قياسيه وحيدة النسيلةالفأري. Abstract

ABSTRACT:BACKGROUND:Lactobacilli are ubiquitous in the environment and in human body. They widely used as probiotic therapeutic agents. However, several authors reported that this bacteria may cause several serious infections in various parts of human body.OBJECTIVE:This work is aimed to illustrate the histopathological effect of lactobacilli in murine spleen which play a major role in the immune system.MATERIALS AND METHODS:Three Lactobacillus isolates (namely, L. bulgaricus, L. plantarum and L. acidophilus) were isolated form yogurt, vinegar and vagina, respectively. These isolates were injected intraperitoneally in mice in a dose 1.5 X 108 CFU/ml.RESULTS:Several histopathological changes were caused by lactobacilli represented by degeneration, necrosis, presence of haemosidrin and macrophages and lymphoblasts.CONCLUSION:This non-pathogenic bacteria was able to cause severe damage to the spleen of mice.

Abstract

Abstract Spleen rupture is a rare life threatening emergency that mostly occurs in pathologically enlarged spleens. Causes may include infections, congenital cyst, metabolic, degenerative and malignancy (leukemia). Several possible mechanisms have been proposed for spleen rupture in patients with hematological disorders. Most commonly, malignant cells of lymphoproliferative or myeloproliferative origin infiltrate the spleen directly. Their sheer volume exceeds the capacity of the relatively non-distensible splenic capsule, causing splenic rupture and splenic hemorrhage.In this report we present a 34 years old male patient presented with an acute abdomen after 3 days from sustaining blunt hit over the abdomen. After immediate resuscitation, exploratory laporotomy had done that revealed old altered blood in the peritoneal cavity and large spleen with non-bleeding laceration in addition to hepatomegaly and dilated mesenteric veins.
