EFFECTS OF DEXAMETHASONE, ESTROGEN ADMINISTRATION ON LEPTIN, THYROID, REPRODUCTIVE HORMONE CONCENTRATION AND LIPID PROFILE OF FEMALE RABBITS SERUM.

Abstract

The aim of this study investigate the effects of long term administration ofDexamethasone (DEX.) 17 β-estradiol (EST.) on body weight, concentration of somehormones ( Leptin, thyroid and reproductive hormone) in addition to determination oflipid profile of female rabbits blood serum.Thirty local rabbits were subdivided randomly and equally into three groups. Thefirst group was used as a control which injected with normal saline . The second groupwas injected daily (0.5mg /kg B.W) DEX . Intramuscularly. The third group was injectedsubcutaneously daily with 0.3 mg /kg B.W s/c 17 β-estradiol. All groups in this studywere treated for 60 days , and blood collected every 15 days. Serum was tested for leptin,LH, FSH, T3, T4 hormones total Cholesterol(TC), HDL(height density lipo-protein),LDL (low density lipo- protein),TG (triglycerides), Body weight was taken every 15 daysfrom the first day .The result showed significant (P≤0.05) decreased in body weight afteradministration of both dexamethasone (DEX.) and Estrogen (EST.), significant increasedin leptin concentration with dexamethasone (DEX.) Treatment while significantdecreased with Estrogen (EST.) treatment, in the other hand LH and FSH concentrationshowed significant decreased in both treatment dexamethasone (DEX.) and Estrogen(EST.) thyroid hormone show significant decreased with dexamethasone (DEX.)Treatment and significant increased T3 & T4 hormone with Estrogen (EST.) treatment ascompare with control while the increase in the T4 it was occurred only in the periods (45and 60)days within treatment groups , lipid profile (TC,TG,LDL, HDL,VLDL) showsignificant increased with DEX. treatment while Significant decreased with EST.treatment

Abstract

Objective: to determine the long-term effect of glibenclamide on thyroid function tests in type 2 diabetic patients. Materials and methods: A total of 63 diabetic patients, 32 type 2 on glibenclamide and 31 type 1 on insulin, were enrolled in this study. Thirty two, apparently healthy volunteers, were also included in the study as a control group. Blood samples were taken from the patients and controls, then the serum were analysed for measurement of fasting serum glucose (FSG), thyroid hormones (total T3, fT3, total T4, fT4) and TSH. Results: the result showed that there were no significant differences between thyroid hormones and TSH of the glibenclamide group when compared to the same parameters in the control group, however the fT3 and fT4 of insulin group were significantly lower than that of the control and glibenclamide groups. There were no significant differences between total T3 and total T4 of control, glibenclamide and insulin groups when compared to each others, whereas the TSH of insulin group was significantly higher than that of control and glibenclamide groups.Conclusion: long term therapy with glibenclamide have no effect on thyroid function tests in type 2 diabetic patients and there was no correlation between glycemic control and thyroid hormones or TSH.

الهدف: لبيان تأثير استخدام عقار الجليبينكلامايد لفترات طويلة على وظائف الغدة الدرقية لدى مرضى السكري من النوع الثاني.الطرق المتبعة والاشخاص: اجريت هذه الدراسة في مدينة الموصل. حيث شملت الدراسة 63 مريضا مصابا بداء السكري, 32 مريضا مصابا بداء السكري من النوع الثاني يتعاطون عقار الجليبينكلامايد و31 مريضا مصابا بداء السكري من النوع الاول يتعاطون الانسولين, و32 شخصا سليما كمجموعة ضبط. اخذت عينات الدم من المرضى ومن مجموعة الضبط وتم تحليل مصل الدم لقياس سكر المصل وفحوصات وظائف الغدة الدرقية وفحص الهرمون المحفز للغدة الدرقية. النتائج: اظهرت النتائج انه لايوجد اي فرق معنوي بين فحص هورمونات الغدة الدرقية كافة وفحص الهرمون المحفز للغدة الدرقية لمجموعة عقار الجليبينكلامايد عند مقارنتها مع مثيلاتها في مجموعة الضبط بينما يوجد فرق معنوي في فحص هورمونات الغدة الدرقية الحرة لمجموعة الانسولين حيث انها اقل من مجموعة الضبط ومجموعة عقار الجليبينكلامايد. لايوجد اي فرق معنوي بين المجاميع كافة عند مقارنة فحص هورمونات الغدة الدرقية الكلية مع بعضها البعض بينما فحص الهرمون المحفز للغدة الدرقية اعلى في مجموعة الانسولين بشكل معنوي عن المجموعتين الاخريتين.الاستنتاج: استنتجت الدراسة ان استخدام الجليبينكلامايد لفترة طويلة لا يؤثر على فحوصات وظائف الغدة الدرقية لدى مرضى السكري من النوع الثاني ولايوجد اي علاقة بين مستوى السكر في الدم وفحوصات الغدة الدرقية.

Abstract

Objective To evaluate the relationship between serum Leptin levels and metabolic syndrome, in (obese, non-obese) control subjects, untreated hypothyroid and hyperthyroid patients. Methods A study was made on 63 goitrous patients and 25 control subjects. Body mass index (BMI) was calculated. Serum T3, T4, TSH were measured by RIA method, serum lipid profile was measured by spectrophotometric method, serum Leptin was measured by ELISA method. Results The mean serum Leptin level was significantly higher in obese than in non-obese control subjects (29.1 vs. 5.8 ngm / ml) and higher in hypothyroid patients (27.9 ngm /ml), and lowest levels in hyperthyroid patients (3.4 ngm /ml). Conclusion Leptin reduce body weight by decreasing food intake (appetite), fat deposition and increasing energy expenditure. Serum Leptin is highly correlated with the body mass index (BMI), other indices of adiposity (lipid profile) in normal obese human and hypothyroid patients.

Abstract

Mature Albino rats of both sexes were given 5mg/kg/day carbimazole orally for 15 days for experimental induction of hyperplasia. The animals divided to four groups, G1 served as a positive control and G2, G3 and G4 treated orally with 50, 100 and 200 mg/kg/day respectively of the aqueous extract of Teucrium polium for 45 days to evaluate its effects on Hb, RBCs, differential leukocytic count and thyroid gland after 15, 30 and 45 days of treatment. The results showed a decrease in Hb and RBCs values during the first two periods and an increase during the last period of the experiment, no significant changes on the differential leukocytic count whereas T4 and T3 revealed a significant increase during the first two periods and a significant decrease during the last period. histopathological preparations of the thyroid gland revealed a remarkable amelioration on the follicular cells in which they were refilled with colloid materials and become look like normal especially in the group treated with 100mg/kg.

عوملت الجرذان البالغة من كلا الجنسين بعقار الكاربمازول 5ملغم/كغم/يوم عن طريق الفم لمدة 15 يوم للتحفيز التجريبي لفرط التنسج . قسمت الحيوانات الى مجموعة سيطرة ايجابية وثلاثة مجاميع عوملت عن طريق الفم بالمستخلص المائي لنبات الجعدة وبجرع 50و 100 و200 ملغم/كغم/يوم لتقييم التاثيرات المحتملة على الهيموغلوبين وكريات الدم الحمراء والحساب التفريقي لكريات الدم البيضاء والغدة الدرقية بعد 15و30 و45 يوم من المعاملة. اظهرت النتائج انخفاض في قيم الهيموغلوبين وتعداد كريات الدم الحمراء خلال الفترتين الاوليتين من التجربة وارتفاع في تلك القيم للفترة الاخيرة من التجربة، لم تظهر تاثيرات تذكر على الحساب التفريقي لكريات الدم البيضاء بينما ارتفعت قيم هورمونات الثايروكسين والثايرونين ثلاثي اليود بشكل ملحوض خلال الفترتين الاوليتين من التجربة وانخفضت تلك القيم خلال الفترة الاخيرة من التجربة. التحضيرات النسيجية للغدة الدرقية اظهرت تحسن ملحوظ للخلايا الحوصلية حيث اعيد امتلائها بالمواد الغروية واصبحت تبدو مشابهة للطبيعية وخاصة في المجموعة المعاملة بجرعة 100ملغم/كغم/يوم.

Evaluation of thyroid function at different stages of pregnancy in Iraqi women.

Abstract

Abstract:The evaluation of thyroid function of either hyperthyroidism or hypothyroidism should beassessed by determination of serum Triiodothyronine (T3),Thyroxin (T4) and Thyroid StimulatingHormone (TSH). Due to specific conditions related to the pregnancy period, there are variousalterations accompanied this stage of life. Some changes required due to physiological demandsof pregnancy. Thyroid function was studied by determination of thyroid hormones using highsensitiveEnzyme Linked Immune sorbent Assay (ELISA) technique in 35 pregnant Iraqi women.The study group comprised 35 full term pregnant women scheduled follows up the alterations ofthyroid hormones, while the control group included 30 healthy women volunteers. Serumconcentrations levels of total T3 (TT3), total T4 (TT4) & (TSH) were estimated using (ELISA)technique. In the study group, blood samples were obtained during various stages of monthlyperiod of pregnancy.Mean age of the study group was (27±5) years, and that of controls were (25±3)years. Infirst trimester: serum TT3 & TT4 levels were significantly higher than that in controls[1.2134±0.0445 vs. 1.0583±0.2439 ng/mL and 8.5266±0.4545 vs 7.0466±1.4460 μ gm/dLrespectively while TSH levels were significantly lower than that of control 2.3866 ±0.3087 vs3.3466±1.3396 μIU/mL; P< 0.05]. In second trimester, there were continuously increase inconcentrations levels of TT3, and TT4 than that in control but TSH significantly was decreased[1.347±0.0191 vs 1.0583±0.2439 ng/ml, 9.5923±0.31005 vs 7.0466±1.4460 μ gm/dL and1.6733±0.1469 vs 3.3466±1.3396 μ IU/mL; P< 0.05]. In third trimester, TT3 showed significantfall [1.2833±0.0447 vs. 1.0583±0.2439 ng/ml and the levels of TT4 significantly increased10.3213±0.0914 vs 7.0466±1.4460 μ gm /dL while TSH significantly decreased1.2685±0.0390 vs3.3466±1.3396 μIU/mL and; P< 0.05]. All alterations, the TT3 in first trimester was risesignificantly, and it was significantly fall in third trimester. TSH was significant fall in thirdtrimester .Conclusion that during pregnancy seemed to be significantly influenced by stresspresent during pregnancy.

الخلاصة:ان تقیم وظیفة الغدة الدرقیة من ناحیة كثرة او قلة افراز ھرمون الثایروكسین یعتمد على قیاس تركیز ال(ثایرونینفي مصل الدم. (TSH) أوالھرمون المحفز للغدة الدرقیة (TT أوالثایروكسین( 4 (TT ثلاثي الیود) ( 3خلال فترة الحمل یوجد الكثیر من التغیرات الوظائفیة حسب متطلبات الحمل، حیث تتأثر وظیفة الغدة الدرقیةاثناءالحمل.في مصل الدم باستعمال جھاز TSH وTT و 3 TT درست وظیفة الغدة الدرفیة اثناء الحمل عن طریق قیاس تركیز ال 4على 35 حامل من النساءالعراقیات. Abstract

Aims: To evaluate the relationship between serum Leptin, Growth factor-1 (IGF-1) levels and untreated thyroid dysfunction patients.Patients and Methods: A study was made on 50 untreated goiterous patients (female and males) with range between (25-55) years; and25 controls subjects (females and males) with range between (14-55) years.Serum T3, T4 &TSH were by Radio immune assay (RIA) method. Serum Leptin and Insulin like growth factor-1 (IGF-1) was measured by ELISA method.Results: The mean Serum Leptin level was significantly lowest in hyperthyroidism than control subjects. While Insulin growth factor level was significantly highest in hyperthyroidism than control subjects (3.82 vs. 2.49)Conclusions: the objective of the study was to provide new data on the relationship between serum Leptin, Insulin like Growth Factor-1 levels and thyroid dysfunction.

Abstract

Gonadotropins (FSH, LH) and testosterone are the prime regulators of germ cell development. Abnormal spermatogenesis is often associated with altered serum gonadotropins and testosterone. the potential relationship between other hormones, including thyroid hormones, and semen quality are still not completely understood. Thus in the present study an attempt has been made to report the degree of associations between thyroid hormones and semen quality. FSH, LH .T. PRL.T4 and TSH.hormones levels were estimated in 39 infertile men of whom 5 were azoospermia, 13 were oligozoospermic, 21 were with asthenzoospermia . Results showed statistically significant differences(P≤ 0.05) in the mean FSH, Testosteron (T.), PRL. And T4. Levels in all the infertile males studied when compared with the fertile controls (n=39). However, there is no significant difference in the mean levels of LH and TSH between the infertile and fertile men. Present study found that though men with abnormal semen profile had slightly higher in T4 concentrations in asthenozoospermia group as compared to those with normal semen profile. This study also show positive correlation between T4 and T. ( R2=0.408) and negative correlation between T4 and FSH was (R2=-0.515) ,T4 and LH was (R2=-0.345).

تعتبر هرمونات المناسل هي منظمات رئيسية للتطور النطف وهي FSH,LH,T. وكذلك مؤخرا لوحظ دور للهرمونات الدرقية ونوعية النطف .في هذه الدراسة تم قياس هرمونات FSHو LHوTو PRLوT4 في 39 من الذكور الذين يعانون من قلة الخصوبة ، وجد 5 منهم يعانون من انعدام النطف و13 يعانون منقلة النطف و21 يعانون من ضعف حركة النطف . أظهرت الدراسة وجود فروق معنوية P<0.05 في معدلات الهرمونات FSH,.T. PRL,T4 مقارنة مع مجموعة السيطرة والتي تضم 39 رجل خصب بينما لا توجد فروقات معنوية P>0.05 في كل من LH وTSH . وجد في الدراسة أيضا وجود علاقة خطية موجبة بين T4 ,و (R2=0.408) T وعلاقة خطية سالبة بين T4وFSH (R2=-0.515) وكذلك بينT4 وLH (R2=-0.345) .

Abstract

Hormones are actual messengers in endocrine signaling. Thyroid gland as endocrine glands holds a critical place in controlling brain and somatic development in embryos and metabolic activities in adults . There is an Coordination between hypothalamus-pituitary-thyroid and hypothalamus-pituitary- gonadal axes, Therefore hypo- and hyper-thyroidism affect women fertility and prolonged exposure to altered thyroid physiology could lead to women infertility . This study was done to illustrate some hormones(sex and thyroid ) changes , the study included 73 infertile women and 23 fertile women24-35years old between March 2013 and June 2013, in Endocrines and diabetes medical center in Nassirhyia city . The results of the study were revealed the significantly increase in the level of LH , FSH , and prolactin and T4 in infertile womens compared with fertile womens(control) while no significant differences was observed in T3 level between two groups .

تعتبر الهرمونات الوسائط نقل الاشاره الكميائيه بين الغدد الصم , وهرمونات الغدة الدرقية تسيطر على عمل الدماغ ونمو الخلايا الجسمية في الاجنة والفعالية الايضيه للخلايا عند الكبار , وهناك تناسق بين هرمونات الغدة الدرقية وهرمونات الغدد التناسلية التي تفرز بايعاز من الغدة النخامية . فالزيادة والنقصان في هرمونات الغدة الدرقية يؤثر في هرمونات الغدد التناسلية . جاءت هذه الدراسة لتحديد التغير الذي يحصل بين هذه الهرمونات ومعرفة هل هناك تاثير لهذا التغير على خصوبة النساء .حيث اجريت الدراسة في المركز الطبي لامراض الغدد الصم والسكري على 73 امراءة تعاني من مشاكل الخصوبه ولعمر من 24-35 ومجموعة سيطرة 23 للفترة مابين اذار 2013 الى حزيران 2013 في مدينة الناصريه واظهرت النتائج ان هناك زيادة في الهرمونات الجنسية LH,FSH ,Pro. والدرقية TSH ,T4.
