Spectrophotometric Determination of Vitamin B6 by Coupling with Diazotized p-Nitroaniline

Abstract

A spectrophotometric method has been described for the determination of pyridoxine hydrochloride (vitamin B6) in pure and pharmaceutical preparations. The method is based on coupling reaction of vitamin B6 with diazotized p-nitroaniline in alkaline solution in the presence of CTAB to form an azo dye which has maximum absorption at 480 nm. Beer's law is obeyed over a range of 5-500g/25ml with a molar absorpitivity of 2.7 × 104 l. mol-1.cm-1 and Sandell sensitivity index of 0.0076 g.cm-2, a relative error +0.22 to + 0.47% and a relative standard deviation of ± 0.11 to ± 0.98%, depending on the concentration level. The proposed method has been applied successfully to the determination of vitamin B6 in pharmactical preparations.

تم اقتراح طريقة طيفية لتقدير البيروكسيدين هيدروكلورايد (الفيتامين B6) بشكله الحر وفي مستحضراته الدوائية. تعتمد الطريقة على تفاعل الاقتران لفيتامين B6 مع 4-نايتروانيلين المؤزوت في وسط قاعدي وبوجود الـ CTAB لتكوين صبغة ازوية تعطي أعلى امتصاص عند 480 نانوميتر. كانت حدود قانون بير 5-500 مايكروغرام / 25مل. وكانت الامتصاصية المولارية 2.7× 104 لتر. مول-1. سم-1 ودلالة ساندل 0.0076 مايكروغرام . سم-2 ، الخطا النسبي يتراوح بين 0.22+ و 0.47 + % وانحراف قياسي نسبي بين ± 0.11 و ± 0.98 % اعتمادا على مستوى التركيز. تم تطبيق الطريقة المقترحة بنجاح في تقدير الفيتامين B6 في مستحضراته الصيدلانية .

Abstract

ackground: Hyperhomocysteinemia has been associated with an increased risk ofatherosclerosis and other cardiovascular diseases. Vitamin B6, vitamin B12, and folicacid are essential components in the metabolism of homocysteine.Objective: This study aimed to assess the effects of vitamin B6, vitamin B12, folic acid, andthe combination of them in the prevention of hyperhomocysteinemia.Methods: Thirty six male local rabbits were used in the study. Methionine was given torabbits to induce a model of hyperhomocysteinemia. Rabbits were divided randomly into 6groups (6 rabbits in each group) as the following: control group: they were maintained onstandard chow only; methionine only group: they were maintained on methionine only; B6group: they were maintained on methionine and vitamin B6; B12 group: they weremaintained on methionine and vitamin B12; folic acid group: they were maintained onmethionine and folic acid; multivitamins group: they were maintained on methionine,vitamin B6, vitamin B12, and folic acid.Results: Multivitamins group showed no significant differences (P>0.05) in homo-cysteinelevels at day 30 in comparison with day 0 and there are no significant differences (P>0.05) inhomocysteine levels at day 30 between multivitamins group and control group. At day 30 andday 60, serum homocysteine levels in multivitamins group were significantly (P<0.05) lowerthan serum homocysteine levels in folic acid group.Conclusion: Multivitamin combination composed of vitamin B6, vitamin B12, and folic acidreduces the hyperhomocysteinemic effect of methionine and it is better than folic acid alone,while folic acid is better than vitamin B6 or vitamin B12 when they used alone.

Abstract

ABED31, 3 Department of Physiology and Pharmacology, College of Veterinary Medicine, University of Basra2 Departments of Animal Production, College of Agriculture, University of BasraAbstractThe present study was undertaken to investigate the role of vitamin B6 in the improvement of the copper sulfate toxicity on some hematological parameters, biochemical parameters and histopathological changes in some internal organs of female rabbits. Eighteen female rabbits were divided to three groups(6 rabbit for each group) .The first group treated with normal saline orally daily for10 days (control),the second group treated with 6mg/kg live weight copper sulfate orally, and third group treated with 6mg/kg live weight copper sulfate orally and7mg/kg live weight vitamin B6 intramuscular injection daily for10 days .The results showed positive effect of vitamin B6 to improvement the different changes which resulted from the exposure to copper sulfate .There was significant increased PCV, Hb concentration and RBC count in third group when compared with second group .Total WBC count percentage improvement and decrease significantly total cholesterol ,ALT and AST in third group when compared with second group .In addition to that the histopathological changes in liver and kidney were return to the normal value with a little alteration as compared with control group.
